- Supercritical Angle Fluorescence (SAF) (dt.: Fluoreszenz oberhalb des kritischen Winkels) ist eine Methode der Mikroskopie zur fluoreszenzspektroskopischen Untersuchung biochemischer Spezies und Strukturen (Proteine, DNA, Zellen, Gewebe). Die getrennte Erfassung der oberhalb des kritischen Winkels abgestrahlten Fluoreszenz ermöglicht die spezifische Untersuchung des Kontaktbereichs der Probe mit dem Tragglas (Objektträger, Deckglas). - La microscopie de fluorescence supercritique (SAF, Supercritical angle fluorescence microscopy) est une technique particulière de microscopie optique à fluorescence permettant l'observation spécifique d'une tranche supérieure ultra fine d'un échantillon (moins de 100 nm d'épaisseur). Cette technique, plus sélective encore que la microscopie de fluorescence par réflexion totale interne (TIRF, limitée à 200 nm), permet de dépasser les limites théoriques que devrait imposer la diffraction de la lumière grâce à une observation dans les angles supercritiques. La méthode a été inventée en 1998 dans les laboratoires de Stefan Seeger à l'université de Ratisbonne en Allemagne et plus tard à l'université de Zurich en Suisse. (fr)
- Supercritical angle fluorescence microscopy (SAF) is a technique to detect and characterize fluorescent species (proteins, biomolecules, pharmaceuticals, etc.) and their behaviour close or even adsorbed or linked at surfaces. The method is able to observe molecules in a distance of less than 100 to 0 nanometer from the surface even in presence of high concentrations of fluorescent species around. Using an aspheric lens for excitation of a sample with laser light, fluorescence emitted by the specimen is collected above the critical angle of total internal reflection selectively and directed by a parabolic optics onto a detector. The method was invented in 1998 in the laboratories of Stefan Seeger at University of Regensburg/Germany and later at University of Zurich/Switzerland. (en)
